The proposed research has as its primary goal to assess the feasibility of microsatellite amplification on a variety of human DNA extracted from archival paraffin-embedded tissues. Since the establishment of the human genome project, several microsatellite polymorphisms have been characterized by researchers who intend to explore genetic aspects of human diseases, but who are limited by the type of sample (e.g., DNA extracted from blood) available. Many of the microsatellite markers are not polymorphic in paraffin-embedded tissue because of differences in the effectiveness of the deparaffinization/rehydration procedure used to isolate the DNA from these tissues. Moreover, paraffin embedding may create additional problems in terms of DNA extraction, especially since the recommended deparaffinization/rehydration procedure sometimes results in the physical degradation of the DNA.
